SGD-01 a new drug for Parkinson’s disease and muscular diseases developed by StaGen.

We have created an explanatory video about SGD-01, a new drug under development by StaGen.
This drug discovery project is led by Naoyuki Kamatani, who previously succeeded in the world’s first gene-based drug discovery. A clinical trial for Parkinson’s disease, supported by AMED, is currently underway at Fujita Health University.

The emergency measures after cardiac arrest have the main purpose of preventing neurological damage due to cerebral ischemia. The Keio University Emergency Medicine Department and others reported that adding hydrogen to inhaled gas may be effective in suppressing neurological damage. Statistical analysis of this trial was conducted by StaGen.

A paper on the anti-aging effect of ATP enhancer published.

A paper co-authored with Prof. Shohei Mitani’s group at Tokyo Women’s Medical University was published. The title is ” Regulation of aging by balancing mitochondrial function and antioxidant levels”.

The paper is on the prevention of aging with XOR inhibitors.

Content Overview.
Administration of febuxostat (FBX), an XOR inhibitor, to C. elegans showed that FBX protects mitochondria and prevents age-related muscle deterioration. Furthermore, the administration of FBX was shown to increase ATP levels and extend lifespan without overloading the mitochondria. Furthermore, we showed that the combination of FBX and antioxidants increased lifespan even more. The study showed that antioxidant effects and increased ATP levels may lead to anti-aging effects.


StaGen’s comment
Febuxostat administration lowers uric acid and may reduce antioxidant function, but the addition of inosine inhibits uric acid lowering. This paper suggests that an ATP-enhancing drug (a combination of febuxostat and inosine) under development by StaGen may have anti-aging applications.

Results of a clinical trial using a new drug [ATP enhancer] sponsored by StaGen Co. Ltd. for the treatment of Parkinson’s disease performed by Professor Hirohisa Watanabe at Fujita Medical School, Professor Emeritus Gen Sobue at Nagoya University, and others, were published online in Medicine (Baltimore). Kamatani, Kamitsuji and Johnson in our company also joined the authors.

In total, administration of [ATP enhancer] to 65 subjects provides support for its safe use in humans, and that latest study in PD patients found that MDS-UPDRS Part III-Motor Score significantly improved after 57 days of treatment. In addition, we were recently granted a patent in the U.S. for our novel method of enhancing cellular ATP and its use as a drug. On top of the previously issued Japanese patent, that patent affords our company protection in two of the world’s most important drug markets. Finally, additional patent applications have recently been published for [ATP enhancer] as an “Anti-aging agent or life extending agent” and as an “Agent to protect mitochondria, ameliorate mitochondrial damage, or improve mitochondrial function”. Those publications allow us to now freely share the underlying data, which we believe represents exciting future growth opportunities.

By analyzing the published data from the CARES study, we found that mortality was higher after discontinuing gout medications allopurinol and febuxostat than during the administration period. For both drugs, the mortality rates were more than 18 times higher within the first month after discontinuation as compared with during the administration period, and the mortality rate gradually declined in about half a year. We considered that both drugs decreased the mortality rate by the ATP enhancement during the administration, but the beneficial effect was abolished by the discontinuation of the drugs. This phenomenon may be called “withdrawal syndrome”. The results of CARES study revealed the mystery that most of the deaths occurred during the off-drug periods, but we think that our present results, for the first time, solved that mystery.
